Effect of high-caloric diet on brain insulin action in mice and humans

Insulin action in brain regulates metabolism and cognitive function. We and others demonstrated that obese and type 2 diabetic mice and persons suffer from brain insulin resistance. Brain insulin resistance contributes to obesity, promoting overeating and unhealthy food choice.
This project will unravel the impact of brain insulin signaling in mice (collaboration with André Kleinridder) and humans after short term high fat diet and the potential impact of this dysregulation on behavior, especially impulsive behavior, attention and food choice.
We will use a high-fat diet to induce acute whole-body insulin resistance in 15 healthy young normal-weight men.
Participants are instructed by a trained nutritionist to increase their caloric intake by 1500 calories a day (50% fat) for one-week. We will investigate peripheral insulin sensitivity and body fat distribution (whole-body magnetic resonance imaging (MRI)). On a separate day, we will use intranasal insulin in fasted participants to assess basal insulin signal in the brain. This will allow us to associate brain insulin sensitivity with behavioral and metabolic alterations.
